Rome would be my recommendation. Loads to see and do. Place is just oozing with history and beautiful buildings. Weather should be decent in October too.
Thought about Prague but know from experience that it's all stag and hen do's which is not a bad thing if your on the trip but not when your there for a romantic/few days break.
Prague has two sides to the city - one with all the bars and stag do etc and one with all the history etc - you can go there and have a romantic time and not see one stag do except st the airport
Never done one of those but I've heard so many good things about them. Available in loads of cities.

Berlin is on my radar to visit this year, as is Prague.

Not sure if these have been mentioned but in recent years I've been to:
Brno - a miniature version of Prague but fewer tourists and cheaper
Bratislava - charming little city often mistakenly overlooked in favour of Vienna
Kaunas
Split
Oslo

But I'd argue to avoid all the stag and hen dos you then have to avoid the Old Town in the evening, which kind of will detract from the visit there wouldn't it?
