I know that if you meet the criteria, a 4BBB card can count for your handicap, but can you mark a card for handicap while playing in a 4BBB?

I was asked this question as "someone who generally knows what what" and I feel you shouldn't but I cannot find anything that appears to forbid it.

Any thoughts?

I'm pretty sure this has come up before and the answer is no. I can't point you towards anything but I'm sure someone will be along shortly to do so.

Rules of Handicapping, Rule 2.1a, Authorised Format of Play. Individual Strokeplay only.

(4BBB scores are only acceptable from organised competitions, and the score has to meet certain criteria).

No, as said it is not an authorised form of play.
Bizarrely almost every time we run a 4BB Open there are a few players who submit GP cards at teh same time. We, of course, delete them and inform their club. I am not sure what they think will happen if they do meet the criteria for an acceptable score in the competition- 2 scores on their record for the same round?

Happens a lot at our Opens. I’m afraid I don’t inform them or their Clubs , I just delete them. (There’s only so many hours in a day!)
No one has ever got back to us to complain.

If the open is all day, you can switch off the tee set(s) on the WHS platform to stop the submission of GP scores for the day

Worth noting that players can (and do) simply resubmit scores through MyEG (or get their club to reenter them manually), so it's better to mark as match play rather than deleting.

The players should be educated to (hopefully) prevent it happening again and to spread the word, so it's important to feed back to their home club (via county if necessary).

I suspect that "advice" is freely exchanged during a round when GP cards are being submitted?
No, no - all GP rounds are all invariably pre registered, properly attested, played off the correct tees and played strictly to the rules of golf (the advice rule in particular is carefully adhered to). This is something that always happens or at least that is what we are told 😉.

No less than in regular club competitions anyway!
